Abstract   Allelic deletions or point mutations of tumor suppressor genes have been considered as an important mechanism during the development of human tumors. APC(adenomatous polyposis coli) and MCC(mutated in colorectal cancer), the tumor suppressor genes, were identified and the cDNA was cloned from chromosome 5q21. Point mutations and loss of heterozygosity(LOH) affecting APC and MCC genes are seen in the hereditary syndrome, familial adenomatous polyposis, and sporadic colon cancer. However, LOH of APC and MCC genes has not been described in gastric cancer and, furthermore, little is known about any tumor suppressor genes that may contribute to the development of gastric cancer. In order to identify frequency of LOH of APC and MCC genes for gastric cancer, we amplified DNA by polymerase chain reaction(PCR) and carried out variable number tandem repeat(VNTR) analysis for MCC gene and restriction fragment length polymorphism(RFLP) for APC gene. We emamined the 45 gastric cancer patients DNAs extracted from paraffin-embedded tissues, including tumor area, gastric mucosa surrounding tumor and normal lymph node. LOH of MCC gene was detected in 77.7%(10 of 13), and especially in 7 cases LOH was also occurred not only in tumor area but also in gastric mucosa area. LOH of APC was less frequent than that of MCC, 33.3%(6 of 18) and allele loss at one or more of MCC and APC loci was detected in 78.3% of 23 informative cases. These data. suggest that the incidence of LOH-affecting MCC was higher than that of APC and, DNA chanse that may be associated with tumor progression was also found in gastric mu- cosa areas. They.imply that MCC is the best candidate for tumor suppressor gene in the development of gastric cancer.
